You mean a zpool on partitions as opposed to the whole raw disk? The only
alignment that is important in this case is to ensure that your pool is
aligned to physical sectors (or hardware RAID blocks if that is what you
use - but don't use that if you have ZFS). This is only an issue if your
disk has physical sectors different from logical sectors. In reality, just
make sure it is aligned to a 4KB boundary to make sure this doesn't cause
you performance issues later if you ever have to replace a disk with a
newer one.

Jumbo ethernet frames have nothing whatsoever to do with clustering.
